He was saying that he has the feed of Bono's vocals in his ear at all times, and Letterman's lapel mic is on the same channel.

During breaks when Letterman takes a break or a guest leaves the set, Letterman makes snide/sly/hilarious comments about the previous guest such as, "Oh, I bet she'll be back" after a particularly bad segment.
