Wow, this is tough. I can just name a few off the top of my head:

Lucy Ricardo
Hawkeye Pierce (M*A*S*H)
Sam Malone/Diane Chambers (I Cheers)

um... drawing a blank. Some shows I prefer the cast, and not one character, like Cheers, Seinfeld,... It's not usually just one person that makes a show. Oh yeah, someone mentioned Murphy Brown Loved the episode when she gave birth
